Does anything have to be done with the filler neck in order to put the honeycomb piece on?

Nope, you won't have to do anything with the filler neck.

Will the original twist on gas cap be OK.?

You should be okay, twist off gas caps were used on alot of Mach 1's from the factory. That being said I like my flip open cap much better than the twist off I used to have, it just looks more finished to me at the honeycomb panel.

I added a Mach 1 honeycomb panel to my car, no problem with the filler neck and the twist-on gas cap still fitted fine. Here's the proof:
